Identification of Synbiotics Conducive to Probiotics Adherence to Intestinal Mucosa Using an In Vitro Caco-2 and HT29-MTX Cell Model

چکیده
The ability of bacteria to adhere the intestinal mucosa is a critical property necessary for long-term colonization tract. This can be highly sensitive presence prebiotics. However, limited data are available in this respect beneficial such as probiotics or resident gut microbiota. We previously demonstrated that prebiotics may decrease adherence several pre- and probiotic combinations. Thus, characterizing interactions between numerous combinations involving different classes crucial identifying new synbiotics. Accordingly, here, we extend our prior analyses evaluate adhesion five lactobacilli, six bifidobacteria, one Escherichia coli strains, commercial promising candidates, together with cariogenic Bifidobacterium dentium strain. As an vitro model, Caco-2 mucin-secreting HT29-MTX cells were co-cultured at 9:1 absence Commercial inulin-type fructooligosaccharide Orafti® GR, P95, galactooligosaccharide-based prebiotic formula Vivinal®, including purified human milk oligosaccharides (HMOs) added into cultivation media sole sugar source (2.5% each). Adherence was tested using microtiter plates evaluated percentage fluorescently labeled present wells after three washes. Consistent prebiotics-mediated enhanced observed only strain E. O83. For remaining HMO P95 GR decreased adherence, reaching statistical significance (p < 0.05) out eight (HMO) 11 strains tested, respectively. Conversely, Vivinal® 12 notably, it significantly attenuated Culture Collection Dairy Microorganisms (CCDM) 318. To knowledge, represents first report on influence HMOs sp. seems used formulation synbiotics, supporting wide range probiotics, especially B. bifidum BBV BBM

In vitro characterization of the adhesive factors of selected probiotics to Caco-2 epithelium cell line
The aim of the present study was to analyse the type of adhesive factors of selected probiotic strains. A large number of lactic acid bacteria with intestinal and dairy origin were collected and assessed for adhesion on Caco-2 cell line. From the best adherent bacteria, four strains were selected for further research: Lactobacillus gasseri G7, L. plantarum F1, L. helveticus AC and L. delbruecki...
متن کاملPotentiation of polarized intestinal Caco-2 cell responsiveness to probiotics complexed with secretory IgA.
The precise mechanisms underlying the interaction between intestinal bacteria and the host epithelium lead to multiple consequences that remain poorly understood at the molecular level. Deciphering such events can provide valuable information as to the mode of action of commensal and probiotic microorganisms in the gastrointestinal environment. Potential roles of such microorganisms along the p...
متن کاملAdherence to and penetration of human intestinal Caco-2 epithelial cell monolayers by Pseudomonas aeruginosa.
Clinical isolates of Pseudomonas aeruginosa from blood adhered to and penetrated intestinal Caco-2 cell monolayers to a greater degree than did isolates from sputum, with a concomitant drastic decrease in transepithelial electrical resistance. PAO-PR1, an avirulent exotoxin A mutant of PAO1, did not cause a decrease in the resistance. The Caco-2 monolayer system may be useful for the evaluation...
متن کاملEffect of edible oils on quercetin, kaempferol and galangin transport and conjugation in the intestinal Caco-2/HT29-MTX co-culture model.
Solubility and matrix play an important role in the gut lumen in delivering bioactive compounds to the absorptive surface of enterocytes. The purpose of this study was to determine the effect of certain commonly consumed lipids, soybean, olive and corn oil, on the transport and conjugation of flavonols (myricetin, quercetin, kaempferol and galangin) using the conjugation-competent co-cultured C...
